A few microliters off can flatten your standard curves, destroy reproducibility, and make you question perfectly good science.<\/span><\/p>\n<\/div><div class=\"fusion-text fusion-text-2\"><p>Here are the <strong>key principles of correct pipette operation<\/strong> (air-displacement pipettes):<\/p>\n<ul>\n<li><strong>Set the volume properly<\/strong>: always approach the desired volume from above (dial down, not up) so the internal mechanism settles correctly.<\/li>\n<li><strong>Fit the tip once, and well<\/strong>: one firm push is enough; over\u2011hammering the pipette into the rack can deform the tip and affect accuracy.<\/li>\n<li><strong>Control angle and immersion<\/strong>: keep the pipette close to vertical when aspirating and immerse the tip only a few millimetres below the surface to avoid extra pressure or air intake.<\/li>\n<li><strong>Pre<\/strong>-wetting the pipette tip<\/li>\n<li><strong>Use the two stops as intended<\/strong>:\n<ul>\n<li>Aspirate only from the <strong>first stop<\/strong>.<\/li>\n<li>When dispensing, press to the <strong>first stop<\/strong> for the set volume, then, wait one second and press to the <strong>second stop<\/strong> to blow out the residual droplet.<\/li>\n<\/ul>\n<\/li>\n<li><strong>Move smoothly and consistently<\/strong>: avoid fast clicks; a steady plunger speed reduces bubbles and variability between replicates.<\/li>\n<li><strong>Protect your samples (and your data)<\/strong>: change tips between samples or reagents, and use filter tips for PCR, DNA\/RNA, cell culture and clinical samples to reduce aerosol contamination.<\/li>\n<li><strong>Look after the pipette<\/strong>: keep it vertical with liquid in the tip, never lying down, and check calibration and cleaning on a regular schedule.<\/li>\n<\/ul>\n<p>The irony is that we invest heavily in sophisticated equipment, yet a <strong>10\u201315 minute refresher on pipetting technique<\/strong> could have a bigger impact on data quality and reproducibility than many new instruments.<\/p>\n<\/div><\/div><\/div><div class=\"fusion-layout-column fusion_builder_column fusion-builder-column-1 fusion_builder_column_1_2 1_2 fusion-flex-column\"
